Abstract:
Objective To study the expression of DNA binding differen-tiated inhibiting protein 1 (Id-1) in human colorectal adenocarcinoma, and explore the role of Id-1 in the pathogenesis of colorectal adenocarci-noma.Methods The samples of colorectal adenocarcinoma(50 cases) and normal colorectal tissues ( 50 cases ) were collected.Reverse tran-scription polymerase chain reaction and Western blot method were used to detect the expressions of Id -1 mRNA and protein in the 50 specimens with colorectal adenocarcinoma and normal colorectal tissues.At the same time, immunohistochemistry was detected to study the distribution of Id -1 protein in colorectal adenocarcinoma and normal colorectal tissues.Results The expression of Id-1 mRNA in colorectal adenocar-cinoma ( 0.96 ±0.03 ) were higher than those in normal colorectal tissues (0.20 ±0.04) ( P<0.01 ).The expression of Id-1 protein in colorectal adenocarcinoma (0.82 ±0.04) were also higher than those in the normal colorectal tissues(0.31 ±0.02) (P<0.01).Pearson correla-tion analysis showed that the Id-1 mRNA level was positive correlation with the protein level ( P <0.01 ).Of the 50 colorectal adenocarcinoma samples ( 72.00%) were positive for Id-1 expression.And of the 50 normal colorectal samples ( 24.00%) cases were positive for Id -1 expression.The positive rate of Id-1 expression in colorectal adenocarcinomasamples was higher than that in the normal colorectal samples ( P<0.01).Conclusion The specific high expression of the Id-1 protein is in a high correlation with the pathogenesis of colorectal adenocarcinoma.
