Effects of triptergium glycosides on the expression of MMP-9 and TIMP-1 in rats with early diabetic nephropathy

Abstract:
Objective To investigate the effect of different doses of triptergium glycosides ( TG) on the expression of MMP-9 and TIMP-1 and the level of interleukin(IL) -6 and C-reactive protein(CRP) in serum in early diabetic nephropathy ( DN ) rats and to explore its mecha-nism of renal protection.Methods A total of 32 SD rats were randomly divided into normal group, diabetic model group, small dose TG group and double dose TG group, 8 rats in each group, and then the data of albumin ( ALB ) , BUN, SCr and 24 hours urinary albumin excretion ( UAE ) and level of IL -6 and CRP after 8 weeks were measured. Expression of MMP -9 and TIMP -1 were detected by immunohisto-chemistry, and protein expression of them in kidney tissue were detected by western blot.Results Compared with normal group, the BUN, UAE and IL-6, CRP in model group were significantly increased (P<0.01). Compared with model group, the ALB, BUN, UAE and IL -6, CRP were better significantly in TG intervention group ( P<0.05 ) , particularly in double dose TG group.Immunohisto-chemistry and western blot results show that compared with normal group, renal tissue expression of MMP-9 model group decreased significantly ( P<0.01 ) , while expressing TIMP-1 increased significantly ( P<0.01 ); Compared with model group, TG intervention group MMP -9 expression gradually increased ( P <0.01 ) , while expressing TIMP-1 expression gradually reduced ( P<0.01 ) , double dose TG group is more obvious.Conclusion MMP-9 and TIMP-1 may be involved in the pathogenesis and progression of DN.TG can ameliorate the renal injury of diabetic rats by reducing the release of IL-6 , CRP and regulation of expression of MMP-9 and TIMP-1 level then improve the matrix deposition.The effects of different doses of TG on DN are different, double dose TG may effect more apparent.
