Abstract:
Objective To develop a simple and rapid high -perfor mance liquid chromatography ( HPLC ) method for detecting the concen-tration of imipenem in critically ill patients.Methods The Alltima HP Hilic column (4.6 mm ×250 mm,5 μm) was used with acetonitrile -ammonium acetate ( 60∶40 ) as the mobile phase .The flow rate was 1 mL? min -1 and the column temperature was 35 ℃.The detector was operated at 298 nm.And then the specificity , lower limit of quantitation and standard curve , precision and recovery rate and stability of the method was investigated.Results Imipenem can be separated well , and the lowest limit of quantification was 0.5 μg? mL-1 .The regressive equation was A =4.9 ×10 3 C -216 ( r =0.999 5 ) , and the linearity range was 0.5-80.0 μg? mL-1 .The intra-day and inter -day RSD were both lower than 15%. The relative recovery rate ranged from 76.17%to 77.42%and the absolute recovery rate ranged from 93.90%to 104.66%.Conclusion This method is rapid, simple,accurate and stable , therefore it is suitable for the detection of imipenem in the human plasma , also can be used for the study on clinical pharmacokinetics of critically ill patients.
