Abstract:
Objective To compare the anti-hepatitis B virus ( HBV) activity between extracted high mobility group chromosal protein N2 ( HMGN2 ) and recombinant HMGN2 , and to explore the anti -HBV effect of HMGN2 molecules.Methods HBV DNA was transferred in the HepG 2.2.15 cell line, and various concentrations of extracted HMGN2 and recombinant HMGN2 were applied to these cells.Superna-tant fluids were collected at day 4 and day 8.ELISA method was used to detect hepatitis B surface antigen ( HBsAg ) and hepatitis Be antigen ( HBeAg) , and real -time PCR method was used to detect the HBV DNA.Results Within 1 to 100 mg· L-1 range, the extracted HMGN2 and recombinant HMGN2 had no cytoxixity in HepG 2.2.15.Within 1 to 5 mg · L-1 range, the extracted HMGN2 and recombinant HMGN2 significantly inhibited the expression of HBeAg and HbsAg, and the HBV DNA copy number was decreased.The antiviral effect was no longer in-creased when the dosese reahced 5 mg· L-1.There was no difference in antiviral effect between the extracted HMGN2 and recombinant HMGN2 when at the same dose.Conclusion Like the extracted HMGN2 , the recombinant HMGN2 also have anti-HBV activity in vitro.
