Abstract: Objective To study the plasma protein binding rate of escin A in rats'plasma.Methods Under three different concentrations, this paper used equilibrium dialysis method to imitate the binding process be-tween escin A and the plasma protein, and determined the concentration of escin A and escin C ( a transformation product of escin A) in and out of the dialysis membrane using high performance liquid chromatography-tandem mass spectrometry ( LC -MS/MS ) method and calculated the plasma protein binding rate of escin A.Results The plasma protein binding rate of escin A in rats was 95.8%, 96.6%and 95.9%at low, middle and high concentration, respectively.There was no significant difference among three concentrations.Conclusion Escin A was highly bound to plasma proteins in rats.
