Effects of contrast agent on neutrophil gelatinase-associated lipocalin in blood and Bax mRNA in kidney of rats with unilateral ureteral obstruction

Abstract:
Objective To study effects of contrast agent on neutrophil gelatinase-associated lipocalin( NGAL) in blood and Bax mRNA in kid-ney of rats with unilateral ureteral obstruction.Methods SD rats were randomly divided into sham,control and contrast agent groups(test).The rats of control and test groups were prepared refolding complete obstruc-tion of the left ureter.The hological changes was deteceted in the kidneys and level of NGAL was measured in rats serum.The bular cell apoptosis was observed and expression of Bax mRNA was measured in kid-neys.Results After the injection of the contrast agent on one day, the volume of rat kidneys, renal parenchymal thickness and weight were sig-nificantly different between left and right kidneys.Compared with control group,the level of NGAL and Bax mRNA in the test group was signifi-cantly higher (P<0.05).The first day and the first three weeks after in-jection, the level of NGAL and Bax mRNA of test group compared with control group was significantly increased ( P<0.05).After the first three weeks,the left renal tubular cell apoptosis in control group compared with test group was significantly higher ( P<0.05 ) .Conclusion Contrast agents have a certain injury in rat kidneys with unilateral hydronephrosis, and apoptosis of renal tubular cell mediated by Bax may be an important mechanism.
