Abstract:
Objective To investigate the protective effects of ginkgo fla-vone aglycone ( GA) on human aortic endothelial cells ( HAECs) apopto-sis induced by oxidized low -density lipoprotein ( ox -LDL ) and its mechanisms .Methods The apoptosis of HAECs was detected by Ho-echst 33258 and AnnexinV -FITC/PI staining by flow cytometry using fluorescence microscope respectively.The gene expressions of P 53 tumor suppressor gene(P53) and B lymphocytes of -2 gene(Bcl-2)were as-sessed by RT -PCR.Results GA inhibited the apoptosis of HAECs markedly.The apoptotic rates were ( 22.35 ± 0.30 )%, ( 16.90 ± 0.36 )%,( 13.66 ± 0.41 )% in HAECs treated with GA at different concentrations , respectively.There was marked difference compared with the injured group ( P<0.01 ).GA can reduce transcriptional level of ap-optosis-associated genes P53 and increased Bcl-2 expression in a con-centration dependent manner after HAECs injured by ox -LDL.Conclusion GA can significantly inhibited ox -LDL -induced HAECs apoptosis , related to inhibition of NF -κB activation signaling pathway, which may be achieved by promoting the expression of Bcl-2, suppress the expression of P53.
