Abstract:
Objective To construct the recombinant expression vector high mobility group N 2 and explore the anti-hepatitis B virus activity of HMGN2 molecule.Methods The pET -32 a-c ( +) -HMGN2 re-combinant expression plasmid was constructed by gene clone technique.His -HMGN2 fusion protein was purified by affinity chromatography.Tricine-SDS -PAGE and Western blotting were used to identify the HMGN2 molecule.The HepG2.2.15 cell line which was infected by HBV DNA was used as a cell model.The effect of anti -HBV and HBeAg in the cell culture supernatant were determined by ELISA.And HBV DNA level in the cell culture supernatant was used to detect by real-time PCR.Results The pET-32 a-c( +) -HMG17 re-combinant expression plasmid was successfully constructed.Those fu-sion protein expression could obviously inhibited the expression of HBeAg and HBsAg;and it decreased the copy number of HBV DNA.Conclusion The HMGN2 in the cell culture supernatant has anti-HBV activity in vitro.
