Effects of carbachol on the visceral proinflammatory factors and dysfunction in rats with sepsis

Abstract:
Objective To investigate effects of carbachol on visceral proinflammatory factors and dysfunction in rats with sepsis.Method One hundred and twenty eight Sprague-dawley rats were projected to a cecal ligation and puncture(CLP)surgery,and divided randomly into two groups:sepsis group and sepsis with carbachol-treatment group (carbachol group).Carbachol(10 μg·kg~(-1))or saline(sepsis group)was injected separately into penis vein immediately after CLP.The mortality rates of 12 h and 24 h after CLP,the levels of proinflammatory factors,the functional parameters and the rates of tissue water content of heart,liver and kidney were examed.Results The mortality rates of 12 h and 24 h of carbaehol group were 25%and 50%respectively;all significantly lower than those of sepsis group 37.5%and 75%.The levels proinflammatory factor,nitrogen monoxide(NO),myeloperoxidase(MPO),as well as the functional parameters and the rates of tissue water content in heart,liver and kidney were significantly lower in carbachol group than those in sepsis group.Conclusion The results indicated that carbochol apparently improve visceral dysfunction and lower mortality rates with its mechanism possibly related to inhibiting the levels of proinflammatory factor and attenuating visceral inflammation and edema in rats with sepsis.
