Abstract:
Objective To investigate the continous inhibitory effect of low dose nimustine (ACNU) , cisplatin(CDDP) and carboplatin(CBP) on the growth of glioblastoma cell line U87 in vitro. Methods U87 cells are divided into 2 groups: One mode is 24 h incubation of U87 with different concentration of ACNU, CDDP and CBP. DMEM was discarded, and incubate for another 24 h or 48 h in fresh DMEM without drugs ( A group). Another mode is U87 incubated with ACNU, CDDP and CBP respectively. The growth inhibition rate of U87 was detected respectively by MTT assay in 24 h, 48 h and 72 h (B group). Results The inhibitory rate of ACNU on the growth of U87 by 2 incubation modes of 48 h and 72 h had no difference (P >0. 05). The inhibitory rate of CDDP and CBP on the growth of U87 by continous incubation group ( B group) is much higher than the group with discard drugs after 24 h (A group) after 48 h and 72 h incubation. The difference of the 2 groups is prominent (P <0.05). Conclusion The inhibitory rate on the growth of U87 in vitro is proportional to the concentration and time of low dose chemotherapy drugs.
