Effects of human adipose-derived mesenchymai stem cells transplantation combined with exercise training on function recovery in rats after spinal cord injury and its related mechanism

Abstract:
Objective To study the effects of transplantation of human adipose-derived mesenchymal stem cells (hADSCs) combined with exercise training on the functional recovery in rats after spinal cord injury(SCI) and the related mechanism.Methods One hundred male SD rats were randomly divided into combination therapy group,cell transplantation group,rehabilitation training group,injury control group and shamed-operated group (n=20).Except for the shamed-operated group,rats in the other groups were induced SCI models using a modified Allen impactor (H=25 mm) at T10.Right after SCI,hADSCs were transplanted into the rats of combination therapy group and cell transplantation group.The combination therapy group and exercise training group started treadmill training on the second day.Hind limb movement was assessed using Basse-Beattie-Bresnaban (BBB) scale before the operation and 1st,2nd,3rd,7th,14th,21th,28th days after SCI.On the 3nd 7th,14th,28th days after SCI,the relative concentration of brain-derived neurotrophic factor (BDNF) of the spinal cord was measured by enzyme-linked immunosorbent assay (ELISA).Results (1)The BBB scores of cell transplantation group and exercise training group were markedly improved since the 7th day of SCI than those in the control group (P<0.05); the scores of the combination therapy group were significantly higher since the 3nd day of SCI than those of the cell transplantation group,rehabilitation training group and injury control group (P<0.05).(2) As compared with that in the shamed-operated group,the BDNF expression in the injury control group decreased significantly since the 3th day of SCI (P<0.05); the BDNF expression in the cell transplantation group and rehabilitation training group were significantly higher than that in the injury control group since the 7th day of SCI (P<0.05),but that in the combination therapy group were significantly higher than that in the cell transplantation group and rehabilitation training group (P<0.05).Conclusions The hADSCs transplantation combined with exercise training have a synergistic effect on functional recovery after SCI,which might be partially due to the up-regulation of BDNF expression.
