Study on white mices with age of 6-8 weeks, 18-20g body weight. They were divided into 3 groups: group 1 (control group): 30 mices; group 2: mices were immunesuppressed by 200mg cyclophosphamid per one kilogram of body weight, being checked after 5 days of injection; and 300mg cyclophosphamid per one kilogram (200mg on the first day and 100mg on the fifth day) and it was checked after 10 days of injection (group 3). The results: immunesuppressed model implemented by intraperitonial injection of 200mg per one kilogram of body weight checked after 5 days of injeciton. Tacrolimus (FK506) is a 23-membered macrolide with immunosuppressant activity that is widely used clinically for treating the rejection after organ transplantation. The research on tacrolimus production was mainly focused on biosynthesis methods, within which there are still some bottlenecks. This review summarizes the progress made in tacrolimus biosynthesis via modification of metabolic pathways and control of fermentation process, with the hope to address the technical bottlenecks for tacrolimus biosynthesis and improve tacrolimus production by fermentation engineering and metabolic engineering.

PURPOSE: Pigs are promising donor species for xenotransplantation. This study was performed to examine acute cyclosporine (CsA) nephrotoxicity in a pig model. METHODS: Adult pigs were treated daily for 1 week with vehicle (VH), or 7.5 mg/kg (CsA7.5), 15 mg/kg (CsA15), and 30 mg/kg (CsA30) CsA. The renal function, electrolyte levels, whole- blood CsA levels, and histopathological results (vacuolization and tubulointerstitial fibrosis) were compared among the different treatment groups. RESULTS: After 1 week of treatment, it was found that CsA induced characteristic lesions that were remarkably similar to those of chronic CsA nephropathy in humans. Compared with the results obtained for the VH group, CsA reduced renal function and yielded poor histopathological results. With an increase in the CsA concentration, the renal function and histological parameters worsened in a dose-dependent manner. CONCLUSION: The study showed that CsA dose-dependently induced renal injury in a pig model. These results provide basic data to estimate or prevent the adverse renal effects of immunosuppressants during porcine xenotransplantation in the future.

Kaposi's sarcoma (KS) is a multifocal, systemic tumor of an endothelial cell origin and there are four clinical subtypes. Among these clinical subtypes, KS associated with immunosuppressive therapy mainly develops in organ transplant patients as well as in other patients who are receiving immunosuppressants for other reasons. It can occur from 1 month to more than 10 years after starting the agent and it seems to depend on the dose and type of the drug taken. We report here on a case of diffuse cutaneous iatrogenic Kaposi's sarcoma that was associated with 2 years of oral corticosteroid therapy. The patient's disease was present as various forms of cutaneous lesions without any systemic involvement.

Necrotizing fasciitis (NF) is an uncommon, life-threatening infection of the subcutaneous tissue and superficial fascia. The clinical course of NF is rapid and progressive, and it is often fatal despite the appropriate therapy. The prognosis of NF depends on a timely diagnosis and then proper treatment. At onset it may not be possible to clearly distinguish NF from minor soft-tissue infections. Although infection are common due to the use of steroids and immunosuppressive agents in patients with systemic lupus erythematous (SLE), it is interesting that NF has rarely been reported with SLE. Here, we present two cases of NF with SLE.
