Acta Universitatis Medicinalis Anhui 2014;(1):115-116,117
To analyze the correlation between fragmented QRS complexes( fQRS) and inflammation in 136 patients with acute coronary syndromes(ACS). Patients with fQRS had increased hs-CRP levels (P<0.01) in comparison to patients with non-fragmented QRS. The higher hs-CRP levels was,the more the number of leads in the fQRS on ECG was(P<0.01). When we performed multiple logistic regression analysis, fQRS was found to be related to in-creased hs-CRP levels [ OR:1.507 , 95% CI:1.172~1.936 , P<0.01 ] . fQRS was independently related to in-creased hs-CRP. fQRS that may result as an end effect of inflammation at cellular level can represent increased car-diac risk in patients with ACS.
2.Anti-hyperuricemic activity of bergenin and its mechanism
Acta Universitatis Medicinalis Anhui 2014;(1):63-66,67
Objective To explore the anti-hyperuricemia activity of bergenin in the model of hyperuricemic mice induced by potassium oxonate. Methods 60 Kunming male mice were divided randomly into six groups, which were normal control group;hyperuricemic model group;and hyperuricemic groups with 20 , 40 , 60 mg/kg berge-nin, and 5 mg/kg allopurinol. Mice were orally administered once daily with 250 mg/kg potassium oxonate for 7 continuous days to create the model, and then three doses of bergenin and allopurinol were orally initiated on the day 1 h after potassium oxonate was given, separately. Serum uric acid, creatinine and urea nitrogon levels, as well as urinary uric acid and creatinine levels were measured. mRNA and protein expression levels of mouse kidney u-rate transporter 1(URAT1), and glucose transporter 9(GLUT9) were also determined. Results Compared with hyperuricemic model group, bergenin significantly reduced serum uric acid, creatinine and urea nitrogon levels, in-creased 24 h uric acid and creatinine excretion, and fractional excretion of uric acid in hyperuricemic mice;mRNA and protein levels of mURAT1 and mGLUT9 were also markedly down-regulated. Conclusion Anti-hyperuricemia effect of bergenin is attributed to the enhancement of uric acid excretion and reversion of mouse urate transporters o-ver-expression.

Acta Universitatis Medicinalis Anhui 2017;52(4):570-574
Objective To investigate the clinical features, BCL-2,CMYC protein expression and prognosis of patients with hepatitis B virus-associated diffuse large B-cell lymphoma (DLBCL).Methods A retrospective study was used to analyse 94 diagnosed DLBCL patients clinical data.Immunohistochemistry method was used to detect the expression of BCL-2 and CMYC protein in paraffin sections of tumor tissues, and to analyze the clinical features, protein expression and prognosis of patients with hepatitis B virus-associated DLBCL.Results ① The rate of HBV infection in the 94 DLBCL patients was 27.66%, significantly higher than the general population(7.18%).Compared with HbsAg-negative DLBCL, HbsAg-positive DLBCL displayed more advanced disease(P=0.032), higher international prognostic index (IPI)(P=0.047) and more frequent involvement of spleen(P=0.02).There were no significant differences in gender distribution, age, immunological subtype and treatment between the two groups.② The aspect of BCL-2 and CMYC protein expression between the two groups: HbsAg-positive group BCL-2 protein expression was significantly higher than HbsAg-negative group (84.6% vs 58.8%, P=0.018).There were no significant differences on the CMYC protein expression and CMYC/ BCL-2 double expression.③ Univariate survival analysis showed that hepatitis B virus infection, BCL-2 positive, CMYC positive, CMYC/ BCL-2 double expression, IPI high-risk group were associated with unfavorable prognostic of overall survival time (OS) and progression free survival (PFS).Cox multivariate analysis showed that CMYC/BCL-2 double expression, IPI high-risk group were independent adverse prognostic factors for OS and PFS.Conclusion HBV infection, BCL-2, CMYC, BCL-2/CMYC double expression, IPI are related to the prognosis of DLBCL.HBV infection may affect the development and prognosis of DLBCL by regulating the expression of BCL-2.

Acta Universitatis Medicinalis Anhui 2017;52(4):475-479
Objective To explore the level of autophagy induced by oxygen glucose deprivation/reperfusion(OGD/R) injury in L02 cell. Methods L02 cells were cultured to establish the model of OGD/R injury and simulate clinical hepatic ischemia-reperfusion injury. The L02 cells were randomly divided into 5 groups : normal control group, oxygen-glucose deprivation 6 h/reperfusion 1,3,6,12 h group (OGD 6 h/R 1,3,6,12 h).Then observe the form changes of the L02 cells by optical microscope. The appreciation of the company's relative L02 cells was detected by MTT.The expression of autophagy related proteins such as Beclin-1, LC3 and p62 were evaluated by Western blot. Results Compared with the normal control group, the form damaged and the cells proliferation activity of L02 cells in the OGD/R group were gradually increased in a time-dependent manner.Compared with the normal control group, autophagy related proteins LC3 , Beclin-1 were increased at OGD 6 h/R 1 h.The expression of LC3 was gradually increased as the time went on and was increased gradually at OGD 6 h/R 6 h, reached a peak at OGD 6 h/R 12 h(P<0.01).The expression of Beclin-1 was gradually increased as the time went on and was increased gradually at OGD 6 h/R 6 h and OGD 6 h/R 12 h (P<0.01).The expression of p62 had no obvious change at OGD 6 h/R 1 h and OGD 6 h/R 3 h, began to increase sharply at OGD 6 h/R 6 h and reached a peak at OGD 6 h/R 12 h(P<0.01).Conclusion Our data suggests that oxygen-glucose deprivation/reperfusion may increase the level of autophagy and lead to autophagic cell death in L02 cell.
