2.Effect of nicotinamide phosphoribosyltransferase on proliferation and apoptosis of human multiple myeloma U266 cells and its mechanism

Objective:To investigate the effect of small interfering RNA (siRNA) silencing nicotinamide phosphoribosyltransferase (NAMPT) gene expression on proliferation and apoptosis of human multiple myeloma U266 cells and its mechanism.Methods:In vitro, NAMPT gene-specific siRNA was synthesized to transfect U266 cells. The experiment was divided into si-NAMPT group (transfected siRNA-NAMPT U266 cells) and si-NC group (transfected negative control siRNA U266 cells). The proliferation of U266 cells after transfection was detected by the methyl thiazolyl tetrazolium (MTT) method, and the apoptosis was detected by flow cytometry. Western blot was used to detect the expression levels of NAMPT, protein kinase B (AKT), phosphorylation-AKT (p-AKT), glycogen synthase kinase-3β (GSK-3β), phosphorylation-GSK-3β (p-GSK-3β), and β-catenin proteins in each group after transfection.Results:Compared with the si-NC group, the proliferation inhibition of U266 cells (absorbance value at 570 nm) increased after transfection for 48 h and 72 h in the si-NAMPT group (48 h: 0.78±0.06 vs. 1.62±0.11; 72 h: 1.23±0.14 vs. 2.37±0.18), and the differences were statistically significant ( t = 3.54, P = 0.034; t = 4.72, P < 0.01). The early apoptotic rate of cells in the si-NAMPT group increased compared with si-NC group [(53.42±0.25)% vs. (25.98±3.18)%], and the difference was statistically significant ( t = 4.41, P < 0.01). Compared with the si-NC group, the levels of p-AKT, p-GSK-3β, NAMPT and β-catenin proteins were significantly reduced in the si-NAMPT group (all P < 0.05). Conclusion:Silence of NAMPT gene can significantly inhibit U266 cell proliferation and induce cell apoptosis, and it may play a role by inhibiting the AKT-GSK-3β-β-catenin signaling pathway.
3.The Treatment with Selective Hepatic Arteriography and Embolization in Liver Hemangioma

Objective We searched for optimal methods of embolization according to the images of selective hepatic arteriography to improve the efficiency of intervention.Methods 40 patients were selected to embolic with lipiodol ultra-fluida and PYM and followed by 6~48 months.Results According its shape and sign of stainangtime,liver hemangioma was distincted from liver cancer.The hemangioma sizes of 30 cases reduced beyond 50 percent and 4 cases had no change.All diminished hemongioma signs can been detected by ultrasound.The effective rate reached 85 percent.All patients had no side-effect.Conclusion The treatment of hepatic hemangioma with selectic arteriography and embolization is a good way in diagnosis and handle.
4.Determination of 2-Chloromethyl-Oxirane in Water by Capillary Gas Chromatography

Objective To make a method for determination of 2-chloromethyl-oxirane in water by gas chromatography. Methods 2-chloromethyl-oxirane in water was extracted by dichloromethane. The extraction was highly condensed.The condensate was isolated through FFAP capillary, and than was detected by hydrogen flame ionization detector. Results The linear range was 0.502-10.04 ?g/ml, r=0.999 2, the detection limit was 0.000 5 mg/ml, recovery rates were 90.3%-99.9%, RSDs were 3.5%-4.3%. Conclusion This method is simple, sensitive, accurate and suitable for determination of trace 2-chloromethyl-oxirane in soak water used for safety assessment of water supply equipments and coatings paints.
5.Relationship Between Chronic Tracheitis and Meteorological Condition and Forecasting Model Established

Objective To set up forecasting models of week and ten days for chronic tracheitis.Methods The data of chronic tracheitis incidence and same time meteorological factors in 1998-2000 in Yinchuan,China were collected and analyzed using SAS6.0 statistical software.Results The incidence of chronic tracheitis was closely related with air temperature,air pressure and the other meteorological factors which showed that chronic tracheitis was caused integrated meteorological factors.Based on the correlation analysis,forecasting models of week and ten days for chronic tracheitis using multi-regression method was established.Conclusion Chronic tracheitis may be caused by integrated meteorological factors and the incidence of chronic tracheitis has a positive correlation with air temperature and a negative correlation with air pressure.
6.Clinical Observation of Recombinant Human Brain Natriuretic Peptide on the Treatment of Acute Decompen-sated Heart Failure

OBJECTIVE:To investigate the effects,prognosis and safery of recombinant human brain natriuretic peptide on the treatment of patients with acute decompensated heart failure. METHODS:90 inpatients with acute decompensated heart failure were divided into observation group and control group according to the use of recombinant human brain natriuretic peptide or not,with 45 cases in each group. Control group received rountine anti-heart failure therapy and intravenous dripping of Nitroglycerin injection at 20 μg/min continuously for 72 h. Observation group was additionally given Lyohilized recombinant human brain natriuretic peptide with initial loading amount of 1.5 μg/kg,iv,then intravenous dropping at 0.007 5 μg/(kg·min)continuously for 72 h,on the basis of control group. Hemodynamic parameters,brain natriuretic peptide level,the difference of liquid intake and output,dyspnea,lung rales,systemic edema,the improvement of cardiac function were observed and compared between 2 groups before and after treat-ment as well as re-hospitalization rate and surriral rate 30 d after medication,the occarrence of ADR. RESULTS:After treatment, left ventricular ejection fraction of observation group was significantly higher than that of control group,and central venous pressure and brain natriuretic peptide level were lower than those of control group,with statistically significance (P<0.05). The difference of liquid intake and output after treatment was (1 171.60 ± 444.80)ml in observation group and (822.30 ± 268.40)ml in control group,with statistical significance between 2 groups(P<0.05). After treatment,the scores of dyspnea,lung rales and systemic ede-ma were decreased in 2 groups;the improvement of observation group was significantly better than that of control group,with statis-tical significance(P<0.05). Total effective rates of observation group and control group were 95.56% and 82.22%;re-hospitaliza-tion rates 30 d after medication were 17.78%and 26.67%;survival rates 30 d after medication were 88.89%and 75.56%,with sta-tistical significance between 2 groups (P<0.05). There was no statistical significanse in the incidence of ADR between 2 groups (P>0.05). CONCLUSIONS:Recombinant human brain natriuretic peptide can significantly improve hemodynamic parameters and heart function of patients with acute decompensated heart failure,and effectively relieve dyspnea,lung rales and systemic edema. It is better than routine treatment in prognosis with good safety.
7.The effect of saponin isolated from stems and leaves of ginseng (SLG) on immunologic function in mice

The effects of SLG on the immune function of normal and immune-suppressed mice were investigated. The results showed that SLG (50mg/kg, im) could correct the suppressive effect of cyclophosphamide, dexamethasone or prednisolone on spleen and. thymus weight, serum specific antibody level and lymphocyte transformation. SLG could also improve the phagocytic activity of peritoneal macrophages and enhance peripheral blood ANAE+-lymphocyte ratio of mice. No obvious effect of SLG on the normal mice were observed except that it could increase the antibody level of normal mice. The results implicate that SLG may have some immunomodulatory effect.

Ⅳ, change of phosphocreatine (PCr) content was very similar to thatof ATP. Content of ADP, AMP, Cr and HYPO was negatively correlated to ATP content. NAD content was also reduced. In summary, recently proposed hyperkalemic crystalloiddilluted blood protecting myocardium would not be sufficient enough and further improve-ment is quiet neccessary.
9.The antagonistic effect of adrenomedullin on extracellular matrix accumulation stimulated by transforming growth factor ?_1 and the cellular mechanism

AIM: To investigate the effect of adrenomedullin on the fibrogenic action of TGF? 1 and the cellular mechanism in renal tubular epithelial cell line (HK-2). METHODS: Total collagen synthesis and secretion were reflected by -proline incorporation and the radioactivity of -hydroproline in the culture medium; ELISA method was used to detect the level of secreted fibronectin (FN); the expression of FN mRNA, collagen IV mRNA and TIMP-1mRNA were determined by RT-PCR; western blot was used to detect the expression of Smad2 and Smad6 proteins. RESULTS: (1) AM could antagonize the expression of collagen and FN stimulated by TGF? 1 in both transcription and translation level; (2) AM down-regulated the TGF? 1-induced TIMP-1 mRNA expression; (3) AM had no effect on the over-expression of Smad2 stimulated by TGF? 1; but could up-regulate the expression of Smad6 inhibited by TGF? 1. CONCLUSION: AM could antagonize the fibrogenic effect of TGF? 1 through up-regulating Smad6 expression in HK-2 cell line.
10.Relationship between Body Mass Index and Serum Prostate-specific Antigen Levels in Prostate Cancer Patients

Objective:The aim of this study was to examine whether PSA levels were affected by BMI and the influence of BMI on the clinical application of PSA.Methods:A total of 856 males who had routine health checkups between January 2006 and July 2007 in our hospital were recruited for this study.Those with a documented history of prostate carcinoma were excluded.Height,weight, and total PSA were measured.The BMl was calculated from measured height and weight [BMl=weight(kg)/height(m)2].Results:The serum PSA(ng/mL)levels were 2.97±0.15 in the normal weight group,2.53±0.12 in the overweight group,and 2.01±0.16 in the obese group(P<0.05).A negative correlation was found between PSA and BMI(r=-0.130,P<0.05)and weight(r=-0.142.P<0.05).A positive correlation was found between PSA and age(r=0.306,P<0.05).Con-clusion:BMl is associated with PSA.As BMI increases, serum PSA decreases.The current data suggest that such an influence should be considered in the clinical application of PSA.
