3.New progress of adoptive immunotherapy in the treatment of hematologic malignancies based on chimeric antigen receptor T-cells: reports from the 56th American Society of Hematology annual meeting
Journal of Leukemia & Lymphoma 2015;24(3):142-146
T cells redirected to specific antigen targets with engineered chimeric antigen receptors (CARs) are emerging as powerful therapies in hematologic malignancies.Various CAR designs, manufacturing processes, and study populations, among other variables, have been studied and reported in clinical trials. The accumulating data supporting CAR-T cells therapy for disease such as chronic lymphocytic leukemia (CLL)and acute lymphocytic leukemia (ALL) highlight what may well become the next sea change in the care of patients with all types of hematologic neoplasia. The scientific symposium on CAR-T cells therapy inspires the mounting enthusiasm regarding this new treatment strategy. Here, the results of the reported clinical trials and the outlook for CAR-T cells therapies were reviewed and discussed. Many questions remain in the field of CAR-T cells directed to hematologic malignancies, but the encouraging response rates pave a wide road for future investigation.
4.5-Azacytidine induces differentiation of bone marrow mesenchymal stem cells into myocardial cells
Chinese Journal of Tissue Engineering Research 2016;20(50):7469-7474
BACKGROUND:Bone marrow mesenchymal stem cel s can be differentiated into myocardial cel s induced by 5-azacytidine in vitro, which provides an opportunity for cel transplantation in the treatment of heart disease. OBJECTIVE:To study the induction effects of 5-azacytidine at different concentrations on the myocardial differentiation of bone marrow mesenchymal stem cel s in rats. METHODS:Passage 3 bone marrow mesechymal stem cel s were incubated with DMEM containing 0, 5, 10, 20μmol/L 5-azacytidine for 24 hours, and then the induced medium was replaced by DMEM containing 5%fetal bovine serum for subsequent 28-day culture. Afterwards, expression of cardiac troponin I was detected by immunocytochemical method. RESULTS AND CONCLUSION:Cel death occurred at 24 hours after 5-azacytidine induction, which was more obvious in the 20μmol/L group than the 5 and 10μmol/L groups. The positive expression of cardiac troponin I was significantly lower in the 5μmol/L group than the 10 and 20μmol/L groups (P<0.05), but there was no significant difference between the 10 and 20μmol/L groups (P>0.05). These experimental findings indicate that 5-azacytidine can induce the differentiation of rat bone marrow mesenchymal stem cel s into myocardial cel s in vitro, and its optimal concentration is 10μmol/L.

Purpose:To further investigate the role of the novel negative immuno-regulatory molecules in tumor evasion by testing B7-H4 e xpression on tumor tissues and spleen macrophages induced by tumor antigen of mi ce and BTLA expression on T cells of the tumor bearing mice. Methods:Via RT-PCR, we evaluated the differences in the expres sion of B7-H4 mRNA of the tumor cell lines (3LL and Renca) in vitro, tumor t issues and the expression of BTLA mRNA of the tumor bearing mice T cells;via flo w cytometry, we evaluated the differences in the expression of B7-H4 surface pr otein of the spleen macrophages from tumor bearing mice and the normal spleen ma crophages after stimulation with the tumor antigen. Moreover, via immunohistoche mistry , we also evaluated the differences in the expression of B7-H4 surface p rotein of tumor tissues. Results:Tumor cell lines (3LL and Renca) did not express the B 7-H4 mRNA in vitro. All tumor tissues or spleen macrophages in tumor bearing mice have a high expression of both B7-H4 mRNA and B7-H4 protein. Compared wi th those from normal mice, T cells from tumor bearing mice showed a higher expre ssion of BTLA. Stimulated by tumor antigen in vitro, normal mice spleen macr ophages B7-H4 expression increased significantly; while no apparent difference was found in the groups stimulated by the corresponding normal tissue antigen. Conclusions:The aberrant expression of the costimulatory molecu les, B7-H4 and its receptor BTLA, may have a regulative effect in tumor evasion immunity.

Successful hematopoietic stem cell transplantation depends on T cells immunol reconstitution. At present, the T-cell receptor excision DNA circles(TRECs)is the reliable index to monitor recent thymic output function. Quantitative analysis of TRECs level can determine the recent thymic output function and evaluate T cells immune reconstitution after transplantation.
