Acta Anatomica Sinica 1954;0(02):-
Objective To investigate the effects of sterigmatocystin(ST) on the ependymal cells, neurons and endothelial cells of blood vessel of the lateral recess of the fourth ventricle in BALB/c mice in vivo after single oral administration. Methods BALB/c mice were treated intragastrically by gavage with ST 3000??m/kg.The mice were killed 1 hour,2 hours,4 hours,8 hours,16 hours after oral administration respectively for observing ependymal cells, neurons and endothelial cells of blood vessel of the lateral recess with transmission electron microscopy(TEM).Results After oral administration of ST pinocytosis vesicles began to increase in endothelial cells of blood vessel in 1 hour, apocrine of ependymal cells began to increase and myelin sheath degeneration appeared in 2 hours, extensive myelin sheath degeneration and mitochondrion degeneration and incisure and vacuoles in nucleus of neuron were found in 4 and 8 hours.Lipofuscin was found in cytoplasm of neuron 16 hour. Conclusion Oral ST exposure in mice could cause myelin sheath degeneration and damaged changes of neurons.

0.05). Conclusions:Compared with normal esophageal mucosa, the expression of HSP27 mRNA in esophageal squamous carcinoma and mucosa with atypical hyperplasia was markedly decreased. This indicated that the expression of HSP27 mRNA to a greater or less extent lost in the carcinogenesis and development of esophageal squamous carcinoma. To up-regulate the expression of HSP27 mRNA in esophageal squamous carcinoma may be a very effective biological therapy.

Objective To explore different intervention methods on the risk factors of cardiovascular disease for women with severe pre-eclampsia history. Methods 78 patients with a history of severe preeclampsia in the hospital from November 2006 to November 2008 were chosen as the research subject. They were divided into the observation group and the control group according to the adopted different methods of intervention with 39 patients in each group. The control group was taken with conventional interventions for obesity intervention, and the observation group was treated with standard behavior therapy (SBT).The improving situations of the related risk factors for cardiovascular disease after three months for the two groups were compared. The effect of different intervention methods on the risk factors of cardiovascular disease was evaluated for women with severe pre-eclampsia history. Results After the intervention, the cardiovascular risk factors (systolic blood pressure, body mass, waist circumference, fasting glucose, total cholesterol/high density lipoprotein) of the two groups improved significantly than before the intervention. But the improvement effect of the observation group was better than the control group, the differences between the two groups were significant. Conclusions The intervention effect on risk factors of cardiovascular disease by standard behavior therapy for women with severe pre-eclampsia history is better than conventional way, which is worthy of clinical application.

Objective To develop a portable jet-flow device for preliminary debridement of wound mainly in battlefield, which can also be used for the treatment of nuclear or chemical weapon injury at wartime and peacetime. Methods The device was composed of a dynamic system,a servo system and consumable accessories, and developed based on a pneumaticpower scheme.Composite power unit free of dependence on electric power transferred the pneumatic power to three kinds of disinfectants through the four-way pipes. Results The device with easy operation consumed a few disinfectant and completed debridement within 30 minutes, with infection avoided effectively. Conclusion The device gains advantages in low weight, compact structure, function and low cost, and can be used for early debridement in field conditions.

ObjectiveTo investigate the influence of Tanshinone Ⅱ A on the expression of HMGB1 in rats with cerebral ischemia-reperfusion (I/R) injury and its neural function protection.MethodsThe 32 male SD rats were randomly (random number) divided into 4 groups (8 rats per group):Sham group,I/R group, group with low dose of Tanshinone Ⅱ A ( TaLD group) and group with high dose of Tanshinone Ⅱ A (TaHD group). The cerebral I/R models were established by the method of right middle cerebral artery occlusion (MCAO).Cerebral infarct volume was detected by TTC staining.Apoptotic cell and apoptotic index were calculated by Tunnel assay. The HMGB1 levels in brain and serum was detected by Western blot and ELISA.Calmodulin (CaM) activity and malondialdehyde (malondiadehyde,MDA) content in the brain were also detected.ResultsCompared with the Sham group, the volume of cerebral infarction, the number of apoptotic cells,CaM activity,MDA content,HMGB1 levels in the brain tissue and serum in group I/R,TaLD group and TaHD group increased significantly (P < 0.01 ).Compared with the group I/R,the volume of cerebral infarction, the number of apoptotic cells,CaM activity,MDA content and the HMGB1 levels in brain tissue and serum in TaLD groupand TaHDgroup decreased significantly (P < 0.01 ).The difference of the above index between TaLD groupand TaHDgroup was significant ( P < 0.01 ).ConclusionsTan Ⅱ A could reduce the cerebral ischemic reperfusion injury in rats which was likely related with decreasing the inflammatory response in the late stage via HMGB1.

Aim To study the effects of amitriptyline(Ami)on focal cerebral ischemia-reperfusion injury in rats. Methods An animal model of focal cerebral ischemia-reperfusion injury was induced by the middle cerebral artery occlusion(MCAO) by reversibly inserting a nylon thread method. The rats were decapitated after ischemia for 1 hour and reperfusion for 2 hours. The infarct volumes were determined using a 2,3,5-tri-phenyl tetrazolium chloride(TTC) staining and assessed by image analysis system. The neurologic deficit status were evaluated on 0~5 grade scale. The levels of dopamine(DA),norepinephrine(NE),serotonin(5-HT) and its metabolic product~hydroxyindole acetic acid(5-HIAA) in cortex and striatum were measured by fluoro-spectrophotometry.Results Ami treatment exhibited a remarkable reduction in infarct volume and neurologic deficit scores. The monoamines content of cortex and striatum had a significant increase compared with ischemia-reperfusion group. Conclusion Amitriptyline has protective effect on cerebral ischemia-reperfusion injury in rats. The mechanism might be related to reducing the release of NE,DA and 5-HT during cerebral ischemia-reperfusion,attenuating or inhibiting of the neurotoxic effects of monoamine neurotransmitters.

Objective To establish a real‐time fluorescent polymerase chain reaction and detect 16S rRNA gene of Legionella strains isolated from sputum specimens of patients with pulmonary infection by using this method .Methods 16s rRNA gene of Le‐gionella was used to design primers and probes .The reaction system and reaction conditions were optimized and the specificity ,sen‐sitivity and repeatability of this method were verified by detecting Legionella pneumophila ,non‐Legionella pneumophila and other bacteria .A total of 557 sputum specimens of patients with pulmonary infection were detected ,and PCR‐digestion identification method was carried out as control .Otherwise ,sequences of 16S rRNA were verified in patients with positive detection results .Re‐sults The results showed that all reference strains of Legionella were positive ,while all of other bacteria were negative ,and the sensitivity was 102 CFU/mL .Among sputum specimens collected from 577 cases of patients with pulmonary infection ,the positive rate of Legionella detected by using real‐time fluorescent PCR and PCR‐digestion identification method was 23 .1% and 19 .9% re‐spectively ,while the positive rate was 17 .2% by verifying the sequences of 16s rRNA .There were no statistically significant differ‐ences of positive rate among the three methods(P>0 .05) .Conclusion The real‐time fluorescent PCR is fast and convenient in de‐tection of Legionella strains isolated from sputum specimens of patients ,which could be an assisted method for clinically diagnosing Legionella infection .
