4.Expression of Echinococcus multilocularis 14-3-3 antigen encoding gene in BCG
Journal of Chongqing Medical University 1986;0(03):-
Objective: To study the expression of Echinococcus multilocularis 14-3-3 antigen encoding gene in Bacillus Calmette-Guerin(BCG).Methods: The recombinant plasmid pBCG-Em14-3-3 was introduced into BCG by electroporation and the bacteria with pBCG-Em14-3-3 were cultured to logarithm growth period, then induced daily by heating at 45℃ for 30min in the last three days. The expression products were analyzed by SDS-PAGE and Western blot. Results: The Em14-3-3 recombinant protein was successfully expressed by pBCG-Em14-3-3 in BCG as an obvious band of 27kDa by SDS-PAGE,which accounted for 11% of total bacterial protein in BCG.Conclusion: The pBCG-Em14-3-3 could be expressed efficiently in BCG and be combined with specific antibody in mice sera, which suggested that the Em14-3-3 recombinant protein expressed by rBCG-Em14-3-3 vaccine was of specific antigenicity.

7.Clinical application MR susceptibility weighted imaging(SWI) in brain diseases
Journal of Chongqing Medical University 2007;0(10):-
Objective:To study the clinical value of susceptibility weighted imaging(SWI)in brain diseases.Methods:37 cases with blood vessel pathological change of brain diseases were collected and analyzed, including 8 example of angioma cavernosum,10 cerebral infarction,15 brain tumor,2 venous malformation and 2 venous sinus thrombus.Conventional T1WI,T2WI,DWI,the SWI and enhanced T1WI,MRA images were performed to compare the superiority of the SWI images to the others in demonstrating the small hemorrhagic focuses, the small vein, hemosiderin, calcification and other paramagnetic materials. Results:SWI could distinguish the hemorrhage from the blood vessel and discover the smaller hemorrhagic focuses of the angioma cavernosum. It could discover more tiny veins adjacent to the dilated draining vein.SWI could detect small hemorrhages in acute cerebral infarction and small draining veins in brain tumor. And draining veins in deep part and expanded rete venosum in the surfaces of brains could be obviously demonstrated when sinus venosus thrombus occurred. Conclusion:SWI is a new pulse sequence that can demonstrate the abnormal low current capacity blood vessels, the structure of small vein, the tiny hemorrhages as well as ferrugination and calcium deposition sensitively. So it can be applied in the diagnosis and antidiastole of the central nervous system diseases as an important supplement to the MRI conventional sequences.
8.Experimental study on rat corneal endothelial damage and repair after acid burn
Journal of Third Military Medical University 2003;0(11):-
Objective To study the effect of acid burn on rat corneal endothelial damage and its repair. Methods The model of mild, moderate and severe acid burn in cornea was established in 90 Long-Evans rats(n=30 for each degree of acid burn) with 4 rats served as normal controls.Corneal edema, opacity and the healing of epithelium were observed with a slit lamp microscope. The corneal endothelium from half rats of each degree acid burn was stained with trypan blue and alizarin red,and that from the other half with hematoxylin and eosin(HE) immediately,30 min,1,3,7,14,21,28 d after cautery. The endothelial damage and its repair were observed and the number of the leucocytes in cornea was calculated under light microscope. Results No damage was seen in the corneal endothelial cells of mild, moderate acid burn, only a few leucocytes could be seen in the cornea, and the healing of epithelium, the regression of edema were within 7 d.The endothelial cells necrosed and shedded in the wound area by severe acid burn. The endothelial cells in the margin were deformed, elongated and slided to the damaged area during repair, and a great number of leucocytes infiltrated, the healing of corneal epithelium was delayed, the regression of edema was up to 28 d.Conclusion The corneal endothelial cells were damaged in severe acid burn. The secondary inflammatory reaction took part in the damage of corneal endothelial cells after severe acid burn. The damage of endothelial cells delayed the healing of corneal ulcer and edema.

Journal of Chongqing Medical University 1986;0(03):-
Objective:To investigate the influences of different OD value and centrifugal parameters on the ultrastructure of Escherichia coli in order to find out the best OD value, speed and time of centrifugation. Methods:According to orthogonal experimental designing principle, the E.coli with different OD600 values are divided into 9 different groups of centrifuge speed and time. The changes of E.coli morphology in each group at different OD600 value, centrifuge time and speed were compared under transmission electron microscope. Results:The different OD600 values and centrifugal parameters have little influence on the ultrastructure of E.coli, but when the OD600 value at 0.8,centrifuge time at 15 min,the cross-section ratio was significantly higher than other groups and was more suitable for observation. Conclusion:The best parameter to prepare the TEM samples of E.coli is under the OD600 value at 0.8 and centrifugation time at 8 minutes and speed at 5 000~12 000 r/min.
10.Development of transcranial magnetic stimulator and measurement of the magnetic field distribution
Chinese Medical Equipment Journal 2004;0(08):-
This paper introduces general guidelines for designing and constructing a magnetic stimulator (MS). These guidelines cover the theoretical concepts, hardware aspects and necessary components. RLC model producing the impulses in MS is focused on and the way to calculate the parameters of the model is presented. Some factors that influence induced magnetic field are discussed. Experiments are carried through with self-developed round coil and 8-shaped, and Tesla meter is used to detect the magnetic fields. The experiments data show that the distribution of the induced magnetic fields by the two coils are tailored to those of theoretical calculation adopted now. A simulation experiment to measure magnetic field in brain has been completed.
