Primary central nervous system lymphoma (PCNSL) is a rare hematological malignancy. PCNSL has the characteristics of strong invasiveness, high recurrence rate and poor prognosis. In recent years, there have been advances in molecular biology research related to the prognosis of PCNSL. This article mainly reviews the research progress in the immunophenotype of PCNSL and the prognosis-related gene mutations, immune checkpoints and copy number variations.

Selection markers are essential tools in gene editing, the utility of such systems is inherently constrained by species-specific limitations, governed by divergent host genetic backgrounds and metabolic compatibility. To address this limitation, we leveraged the CRISPR/Cas9 system to develop a universal counter-selection tool. We designed and introduced an sgRNA expression cassettes as counter-selection markers, which directs the Cas9 protein to target and cleave genomic DNA, allowing for the selection of the strains where the sgRNA expression cassette has been replaced. Optimized to target multiple copy sites with sgRNA, this system significantly enhances cell lethality, boosting counter-selection efficiency to over 85.00%. This counter-selection tool is not limited to single strains and is suitable for various scenarios, including multi-copy plasmid assembly and plasmid editing, demonstrating broad application potential.

Objective:To explore the therapeutic mechanism of moxibustion in Crohn disease(CD)-associated intestinal fibrosis by observing its effects on the angiotensin-converting enzyme(ACE)/angiotensin Ⅱ(Ang Ⅱ)/angiotensin Ⅱ type 1 receptor(AT1R)axis in CD mouse models.Methods:Six randomly selected male C57BL/6 mice were assigned to a normal group,while the remaining mice were administered 0.1 mL of 2,4,6-trinitrobenzene sulfonic acid via enema to establish a CD intestinal fibrosis model.After successful modeling,the mice were randomly divided into a model group,a moxibustion group,and a Western medication group,with 6 rats in each group.The normal group and the model group only received grabbing without intervention.In the moxibustion group,mild moxibustion was applied to Qihai(CV6)and bilateral Tianshu(ST25)once a day for 10 min each time over 7 consecutive days.The Western medication group was administered mesalazine suspension via oral gavage once a day for 7 consecutive days.At the end of the intervention,the general condition,disease activity index(DAI)score,and gross colon score of mice in each group were evaluated.Hematoxylin-eosin staining was used to observe and score the histological changes in the colon tissue in each group.Masson staining was used to observe colonic fibrosis and the ratio of collagen-positive areas was analyzed;the expression of Ang Ⅱ in the colon tissue was detected by the enzyme-linked immunosorbent assay;immunohistochemistry and real-time quantitative reverse transcription polymerase chain reaction were used to detect the protein and mRNA expression of ACE and AT1R in the colon tissue,respectively;Western blotting was used to detect the expression of transforming growth factor(TGF)-β1 and connective tissue growth factor(CTGF)in the colon tissue.Results:Compared to the normal group,the DAI score,gross colon score,colonic histological score,collagen-positive area ratio,ACE protein and mRNA,Ang Ⅱ protein,AT1R protein and mRNA,TGF-β1 protein,and CTGF protein in the colon tissue in the model group increased significantly(P<0.01).In contrast,the above indicators in both the moxibustion group and the Western medication group reduced significantly compared to the model group(P<0.01 or P<0.05).There was no statistical difference in these indicators between the moxibustion group and the Western medication group(P>0.05).Conclusion:Moxibustion can alleviate intestinal fibrosis in CD mice,and its therapeutic mechanism may be associated with the regulation of colonic ACE/AngⅡ/AT1R axis.

ObjectiveTo investigate the efficacy and safety of Shengxiantang in the treatment of early Parkinson's disease with autonomic dysfunction of Qi deficiency pattern. MethodA total of 82 eligible patients were randomized into control group （41 cases） and traditional Chinese medicine （TCM） group （41 cases）. On the basis of standardized treatment of western medicine， TCM group was prescribed Shengxiantang while control group were treated with placebo for 12 consecutive weeks additionally. Scale for Outcomes in Parkinson's Disease-Autonomic （SCOPA-AUT） questionnaire， Unified Parkinson's Disease Rating Scale （UPDRS）， Traditional Chinese Medicine Qi Deficiency Symptom Score of Parkinson's Disease （TCMQDSSPD）， serum levels of glutathione peroxidase （GPx） and superoxide dismutase （SOD）， daily dosage of pramipexole and levodopa and benserazide hydrochloride， and safety index were evaluated both before and after treatment. ResultAfter treatment， the total score of SCOPA-AUT， gastrointestinal score， urinary score， and thermoregulatory score in the control group were higher than those before treatment （P<0.01）， while the above sores in the TCM group were lower than those before treatment （P<0.05， P<0.01）. In addition， the cardiovascular score， pupillomotor score， and sexual score in two groups showed no significant difference from those before treatment. After treatment， the total score of SCOPA-AUT， gastrointestinal score， urinary score， and thermoregulatory score in the TCM group were lower than those in the control group （P<0.05， P<0.01）， and cardiovascular score， pupillomotor score， and sexual score showed no significant difference between two groups. After treatment， the total score of TCMQDSSPD， main symptom scores， and minor symptom scores in the control group had no significant difference from those before treatment. The total score of TCMQDSSPD and minor symptom scores of TCM group were lower than those before treatment （P<0.01）， while the main symptom scores of the TCM group showed no significant difference from those before treatment. After treatment， the UPDRS score， serum GPx and SOD levels， and daily dosage of pramipexole and levodopa and benserazide hydrochloride demonstrated no significant difference from those before treatment in the two groups and between the two groups. No abnormality was found in the safety indexes. ConclusionBased on the standardized treatment of western medicine， Shengxiantang can effectively and safely improve the autonomic symptoms of gastrointestinal system， urinary system， and thermoregulation， as well as the symptoms of Qi deficiency syndrome in early Parkinson's disease with autonomic dysfunction.

Although the development of COVID-19 vaccines has been a remarkable success, the heterogeneous individual antibody generation and decline over time are unknown and still hard to predict. In this study, blood samples were collected from 163 participants who next received two doses of an inactivated COVID-19 vaccine (CoronaVac®) at a 28-day interval. Using TMT-based proteomics, we identified 1,715 serum and 7,342 peripheral blood mononuclear cells (PBMCs) proteins. We proposed two sets of potential biomarkers (seven from serum, five from PBMCs) at baseline using machine learning, and predicted the individual seropositivity 57 days after vaccination (AUC = 0.87). Based on the four PBMC's potential biomarkers, we predicted the antibody persistence until 180 days after vaccination (AUC = 0.79). Our data highlighted characteristic hematological host responses, including altered lymphocyte migration regulation, neutrophil degranulation, and humoral immune response. This study proposed potential blood-derived protein biomarkers before vaccination for predicting heterogeneous antibody generation and decline after COVID-19 vaccination, shedding light on immunization mechanisms and individual booster shot planning.

Medical record information is the core data source for diagnosis related groups(DRG)payment, and high-quality of medical record information is the foundation for DRG payment. The development of medical record management towards medical record information management, pushes up the demand for high-quality medical record information management professionals in hospitals. The conventional practice of professional training had resulted in such setbacks as professionals mismatch, long pre-employment training period, and large demand for continuing education. Based on the triple helix model theory, the study established a production-education integration training mode of university-government-hospital for medical record information management professionals. Capacity development was set as the goal and job demand as the guidance, for effectively integrating the educational resources, and promoting the model of " posts linked to training, employment following graduation" production-education integration training. The aim is to expand the quantity of talent team and improve the quality of talent training, thus effectively easing the shortage of medical record information management professionals.

Objective:To explore effects of family aerobic exercise combined with respiratory training on pulmonary function, self-care ability and quality of life in chronic obstructive pulmonary disease (COPD) patients with frailty.Methods:A total of 86 COPD patients with frailty who were treated in the Outpatient Department in the First Affiliated Hospital of Zhengzhou University from September 2018 to March 2019 were selected by the convenient sampling method. They were randomly divided into the control group ( n=43) and the observation group ( n=43) . The control group was given routine health education while the observation group was given family aerobic exercise combined with respiratory training guidance. After 3 months of intervention, pulmonary function meters were used to compare lung function indexes between two groups. Barthel index and COPD Assessment Test (CAT) were used to compare activity of daily living (ADL) and quality of life of patients. Results:38 patients in the observation group and 40 patients in the control group completed the study. After 3 months of intervention, there was no statistically significant difference in lung function indexes between the observation group and the control group ( P> 0.05) . After 3 months of intervention, the Barthel index score of the observation group (71.28±10.02) was higher than that of the control group while CAT score (13.68±5.16) was lower than that of the control group. The differences between the two groups were statistically significant ( t=-2.102, 2.736; P< 0.05) . Conclusions:Family aerobic exercise combined with respiratory training can improve the lung function of COPD patients with frailty and improve their self-care ability and quality of life, which has certain clinical application values.

Objective? To investigate the effect of stage cognitive behavioral intervention on anxiety and quality of life in patients with esophageal cancer undergoing chemotherapy. Methods? By convenience sampling, totally 78 patients with esophageal cancer combined with anxiety who received chemotherapy in the First Affiliated Hospital of Zhengzhou University from June 2017 to June 2018 were selected as the study subjects. Among them, 40 patients admitted from June to December 2017 were taken as the control group and the other 38 patients from January to June 2018 were in the observation group. The control group was given conventional nursing, and the observation group was given cognitive behavior intervention at admission and within 2 weeks after discharge. The two groups were compared in terms of the anxiety score and quality of life before and after intervention. Results? A total of 36 cases in the control group and 32 cases in the observation group completed the study. Before intervention, there was no statistical difference between the two groups in the anxiety score (P＞0.05). After intervention, the anxiety scores in both groups decreased compared with those before intervention with statistical differences (P＜0.01); and the anxiety score of the observation group after intervention was (40.28±5.46), lower than (55.97±3.28)of the control group with statistical significance (P＜0.01). Before intervention, there was no significant difference in the total scores of quality of life and scores in each fields between the two groups (P＞0.05). After intervention, the score of social/family status field in the observation group was (18.68±6.27) and the total score was (69.85±6.96), both higher than those in the control group (P＜ 0.01), while the scores of physiological, emotional and functional status in the observation group were higher than those in the control group, but there was no statistically significant difference between the two groups (P＞0.05). Conclusions? Stage cognitive behavioral intervention can improve anxiety and quality of life in patients with esophageal cancer undergoing chemotherapy, which is worthy of clinical application.

Objective? To explore the influence of group education based on transactional analysis model on self-management ability and quality of life of patients with chronic obstructive pulmonary disease (COPD). Methods? By convenience sampling, COPD patients admitted in the First Affiliated Hospital of Zhengzhou University from January of 2017 to May of 2018 were selected as research objects and among them, 42 patients admitted from January to August of 2017 were taken as the control group and were given routine health education; 47 patients admitted from September of 2017 to May of 2018 were taken as the observation group and were given group health education underpinned by the transactional analysis model. The COPD patients were compared before and after intervention in terms of the score in Self-management Scale and COPD Assessment Test (CAT). Results? Before intervention, there was no statistical difference between the COPD patients in the two groups in terms of the scores in Self-management Scale and CAT (P＜ 0.05). After intervention, the scores of each dimension of COPD Self-management Scale in the observation group were higher than those in the control group, and the scores of CAT in the observation group were lower than those in the control group, with statistical significance (P＜0.05). Conclusions? Transactional analysis model is conducive to improving the COPD patients' self-management ability and their quality of life.
