1.Clinical analysis of retinal nerve fiber layer thickness and macular fovea in hyperopia children with anisometropia amblyopia

AIM:To analyze the clinical significance of axial length, diopter and retinal nerve fiber layer thickness in hyperopia children with anisometropia amblyopia. ·METHODS: From January 2015 to January 2017 in our hospital for treatment, 103 cases, all unilateral, were diagnosed as hyperopia anisometropia amblyopia. The eyes with amblyopia were as experimental group ( 103 eyes), another normal eye as control group (103 eyes). We took the detection with axial length, refraction, foveal thickness, corrected visual acuity, diopter and the average thickness of retinal nerve fiber layer. ·RESULTS: Differences in axial length and diopter and corrected visual acuity were statistically significant between the two groups (P<0. 05). The two groups had no significant difference in retinal nerve fiber layer thickness (P>0. 05). There was statistical significance difference on the foveal thickness (P<0. 05). There was a negative correction correlation between the visual acuity, axial length with foveal and peripapillary nerve fiber layer thickness, but no significant difference in correlation ( P>0. 05 ). The positive correlation between diopter with nerve fiber layer thickness of foveal and around the optic disc were no statistically significant difference (P>0. 05). ·CONCLUSION:Retinal thickness of the fovea in the eye with hyperopic anisometropia amblyopia were thicker than those in normal eyes;the nerve fiber layer of around the optic disc was not significantly different between the amblyopic eyes and contralateral eyes. The refraction and axial length had no significant correlation with optic nerve fiber layer and macular foveal thickness.
2.Uncertainty Evaluation of Content Determination of Benzoic Acid

Objective:To establish a method for the uncertainty evaluation of the determination of benzoic acid. Methods: The content of benzoic acid was determined by acid-base titration. By constructed mathematics model, the source of the measurement uncer-tainty was analyzed, and the uncertainty components were quantized and combined. Results:The expanded uncertainty of benzoic acid was 0. 36% and the results were expressed as(99. 99 ± 0. 36%,k=2). Conclusion:The mathematics model is reasonable and relia-ble,and can be used in the uncertainty evaluation of content measurement of benzoic acid.
3.Application of Seminar method in teaching Introduction of Nursing Science for advanced vocational nursing students

Objective To explore the effect of seminar applied in instruction of Introduction of Nursing Science for advanced vocational nursing students.Methods A total of 246 advanced vocational nursing students admitted in 2013 from class 1 to class 5 were named as the control group,who received traditional teaching method.Meanwhile a total of 252 advanced vocational nursing students admitted in 2013 from class 6 to 10 were set as the experimental group,and were given seminar teaching.We adopted California Critical Thinking Dispositions Inventory Scale,Academic Self-efficacy Scale and an anonymous questionnaire for teaching satisfaction.Theoretical examination was used to be final evaluation.Results The scores of theoretical examination in the experimental group were higher than that of the control group.The scores of critical thinking ability and academic self-efficacy of the experimental group were higher than those of the control group,with a significant difference.The result of the anonymous questionnaire showed that the experimental group showed a higher satisfaction degree with seminar teaching method.Conclusions Seminar teaching method applied in Introduction of Nursing Science is feasible and effective,which is better than traditional method in improving critical thinking ability,academic self-efficacy and teaching satisfaction.
4.Curative effect of antibiotic de-escalation therapy on severe pneumonia

Objective:To investigate the clinical efficacy and safety of antibiotic de-escalation therapy in the treatment of severe pneumonia.Methods:Sixty patients with severe pneumonia who received treatment in The First People's Hospital of Yongkang,China from January 2015 to January 2018 were included in this study.They were randomly assigned to receive either conventional antibiotic therapy (control group, n = 30) or antibiotic de-escalation therapy (observation group, n = 30).The clinical efficacy,symptom relief time,antibiotic treatment time,length of hospital stay,pulmonary ventilation function index,serum levels of inflammatory factors and the incidence of adverse reactions were compared between the control and observation groups. Results:Total effective rate in the observation group was significantly higher than that in the control group [93.33% (28/30) vs.73.33% (22/30), χ2 = 4.320, P < 0.05].The time to disappearance of cough,fever and abnormal lung sounds in the observation group was (2.10 ± 1.25) d,(2.19 ± 1.24) d,(2.01 ± 0.56) d,respectively,which was significantly shorter than that in the control group [(3.73 ± 1.92) d,(3.68 ± 1.70) d,(3.36 ± 0.78) d, t = 3.897,3.878,7.701,all P < 0.05].The antibiotic treatment time and length of hospital stay in the observation group were (2.89 ± 1.06) d and (4.08 ± 1.23) d,respectively,which were shorter those in the control group [(4.27 ± 1.45) d and (5.76 ± 1.69) d, t = 4.208 and 4.402,both P < 0.05].The forced expiratory volume in one second (FEV 1) and the ratio of FEV 1 to forced vital capacity (FVC) in the observation group were (2.09 ± 0.69) L and (58.94 ± 15.67)%,respectively,which were significantly higher than those in the control group [(1.43 ± 0.57) L,(43.12 ± 11.03)%, t = 4.039 and 4.522,both P < 0.05).Serum levels of C-reactive protein,interleukin-6 and procalcitonin in the observation group were (5.84 ± 2.09) mg/L,(20.05 ± 2.76) ng/L,(2.18 ± 0.78) ng/L respectively,which were significantly lower than those in the control group [(8.50 ± 2.67) mg/L,(23.24 ± 3.07) ng/L,(3.11 ± 0.97) ng/L, t = 4.297,4.232,4.092,all P < 0.05].There was no significant difference in the incidence of adverse reactions between the observation and control groups (6.67% vs.10.00%, χ2 = 0.218, P > 0.05). Conclusion:Antibiotic de-escalation therapy for the treatment of severe pneumonia can shorten the time of disappearance of clinical symptoms,improve pulmonary ventilation function,and inhibit systemic inflammatory reaction without increasing adverse reactions.
5.Optimization of culture conditions of SARS-CoV-2 four Omicron variants in Vero cells

Objective To optimize the culture conditions of four vaccine candidates of severe acute respiratory symptom coronavirus 2(SARS-CoV-2) Omicron variants BA.1,BA.1.1,BA.2 and BA.5 in Vero cells.Methods The harvest time(24,48,72 and 96 h) and MOI(0.01,0.001,0.0001 and 0.000 01) of four Omicron variants cultured in Vero cells were optimized by using cytopathic effect(CPE),viral nucleic acid copy number and viral titer as evaluation indexes.Results The optimum harvest time of the four Omicron variants BA.1,BA.1.1,BA.2 and BA.5 in Vero cells was 72 h,and the optimum MOI was 0.001~0.000 01,0.001~0.000 01,0.01~0.000 01 and 0.01~0.000 01,respectively.Conclusion The culture conditions of four Omicron variants in Vero cells were optimized,which laid a foundation of the development of SARS-CoV-2 Omicron variant inactivated vaccine based on Vero cells.
6.Effect of inhibition of p53 pathway by FAM84B on proliferation of esophageal squamous cell carcinoma cells

Objective To study the expression of FAM84B in esophageal squamous cell carcinoma(ESCC) and its regulatory mechanism on cell growth by p53 pathway.Methods A total of 508 ESCC tumor samples and their adjacent normal tissue samples were collected from Shanxi Cancer Hospital and Affiliated Cancer Hospital of Xinjiang Medical University,which are high incidence areas of ESCC in China.Using whole genome sequencing(WGS) and whole exome sequencing(WES),FAM84B gene with significantly amplified copy number were screened.In ESCC cell line KYSE150with high expression of FAM84B,the effect of FAM84B on cell growth was detected by MTT and hard clone formation assay after interfering with FAM84B by small interfering RNA infection;In ESCC cell line KYSE450 with low expression of FAM84B,FAM84B was overexpressed with plasmid pCMV-3 ×flag-FAM84B,and the effect of FAM84B on cell growth was detected by MTT and hard clone formation assay.The effects of FAM84B knock-down on the expression of CDK4,CDK6and CCND1 were detected by Western blot.Results WGS analysis of 508 ESCC paraffin sections showed 109 cases of FAM84B copy number amplification(21.45%);FAM84B gene existed near 8q24.21 and was a significantly enlarged lesion peak.Knockdown of FAM84B inhibited the proliferation of ESCC cells,while overexpression of FAM84B promoted that.The low expression of FAM84B promoted the cell cycle progression mediated by p53.Conclusion FAM84B can promote cell proliferation by promoting the cell cycle transition from G1 phase to S phase via inhibiting the expression of p53 pathway proteins in ESCC cells.
7.Effect of negative regulation of nucleophosmin 1 by long non-coding RNA growth arrest specific transcript 5 on cisplatin resistance of gastric cancer cells

Objective To investigate the effects of long non-coding RNA(LncRNA) growth arrest specific transcript 5(GAS5) negatively regulating nucleophosmin 1(NPM1) on cisplatin(DDP) resistance of gastric cancer cells.Methods The normal human gastric mucosa cell line GES-1 and human gastric cancer cell lines BG3-823,MGC-803 and AGS were selected as the research objects,of which the level of LncRNA GAS5 in each cell was measured by qRT-PCR.The drug resistance of AGS cells to DDP(AGS/DDP) was induced,and the experiment was divided into control group,empty plasmid group(BC group),GAS5 nonsense interference group(pLJM-GAS5 NC group) and GAS5 overexpression group(pLJM-GAS5 group).MTT method was used to determine the effect of DDP on the proliferation of AGS and AGS/DDP cells;and the levels of NPM1,multidrug resistance 1(MDR1),excision repair cross complementation group 1(ERCC1),multidrug resistance-associated protein 1(MRP1) and N-cadherin in AGS and AGS/DDP cells were measured by Western blot.Results Compared with the normal gastric mucosa GES-1 cells,the level of LncRNA GAS5 in BG3-823 and AGS cells decreased significantly,and among them,the level of LncRNA GAS5 in AGS cells was the lowest,so AGS cells were used for the follow-up experiments.Compared with the control group,the level of LncRNA GAS5 in AGS cells of BC group and pLJM-GAS5 NC group decreased significantly,while the levels of NPM1,MDRl,ERCC1,MRP1 and N-cadherin increased significantly;compared with BC group and pLJM-GAS5 NC group,the level of LncRNA GAS5 in AGS/DDP cells of pLJM-GAS5 group increased significantly,while the levels of NPM1,MDR1,ERCC1,MRP1 and N-cadherin decreased significantly;after treatment with DDP of the same concentration(except 0 μmol/L),compared with the control group,the inhibition rate of AGS/DDP cell proliferation in BC group and pLJM-GAS5 NC group decreased significantly;compared with BC group and pLJM-GAS5 NC group,the inhibition rate of AGS/DDP cell proliferation in pLJM-GAS5group was significantly higher.The semi inhibitory concentration(IC_(50)) of DDP on AGS/DDP cells in pLJM-GAS5 group for 48 h was(65.38±5.04) μmol/L,which was significantly lower than(120.74±4.17) μmol/L and(120.24±4.29) μmol/L in BC group and pLJM-GAS5 NC group.Conclusion Up-regulating the level of LncRNA GAS5 in AGS/DDP cells can reverse the drug resistance of AGS/DDP cells,which may be related to the down-regulation of NPM1expression

Objective Discusses TT3,FT3,TT4,FT4 and TSH the value which examines in the thyroid gland disease patient;analysis thyroid gland disease patient TT3,FT3,TT4,FT4 and TSH change situation.Methods analyzes my hospital 200 example thyroid gland disease patients,and selects the normal adult 50 examples to take the control group.analytic method to each group of examination thyroid function,and carries on the comparison to the group various targets'examination value.Results primary hyperthyroidism group T3,T4,FT3,FT4 were higher than normally,the hypothyroidism group T3,T4,Fr3,FT4 to be lower than normally.The TSH content primary hyperthyroidism group was lower than normally,the hypothyroidism group to be higher than normally obviously.Compares its difference with the control group to have statistics significance.hyperthyroidism group its diagnosis coincidence rate FT3 was 96%,TSH was 96%,T3 was 92%,FT4 was 90%,T4 was 88%.The hypothyroidism,the diagnosis coincidence rate respectively was:TSH was 100%,FT4 was 93%,T4 was 90%,FT3 was 77%,T3 was 70%.Conclusion FT3,T3,TSH the significance are big in the hyperthyroidism diagnosis;FT4,T4,TSH the significance are big in the hypothyroidism diagnosis.
10.Cavolin-1 and thyroid carcinoma

Caveolin-1 is a prominent member of the caveolin family.Caveolin-1 may have closely correlation with some carcinoma' s origination,development,infihration,transference and drug tolerance of tumor cell.But this function has its specificity of cell type and organ.Caveolin-1 can have different functions in formation and progress of tumor cell.Because different thyroid tumors have different structures and growth patterns ,as well as different biological behavior.Expressions of Caveolin-1 have significant differences in thyroid carcinoma with different histological type and differentiation grade.Research on expression of caveolin-1 in different types of thyroid carcinomas and its physiological characteristics play roles in evaluating prognosis of patient with thyroid carcinoma.
