1.Study on effect of vimentin glycosylated mutant plasmid on differentiation of PC12 cells

Objective To construct a vimentin glycosylated mutant plasmid,and to study its impact on rat adrenal chromaffin cell (PC12) cell differentiation.Methods The vimentin glycosylated mutant plasmid was constructed,and the gel electrophoretic was performed and the sequencing identification was performed. The empty plasmid pcDNA3.1b (control group),vimentin plasmid (Vim group) and vimentin glycosylated mutant plasmid (Vim mut group) were used to transfect PC12 cells,which were cultured for 0,3 d in the con-dictions of treatment and non-treatment of exogenous oligosaccharides (Cyclo-ManN pro) respectively,and the cell differentiation records and quantitative analysis (the cell average neurite length and differentiated neu-rite cells percentage) were performed.Results A recombinant plasmid containing a glycosylation site muta-tion site 19(T-G),97(A-G),100(T-G) was successfully constructed (about 1404 bp in size).Before and after Cyclo-ManN pro treatment,the average neurite length of PC12 cells and the percentage of differentiated neu-rite cells in the Vim group were greater than those in the control group and the Vim mut group[(61.98±19.03)μm vs.(51.09±14.45)μm,(51.49±14.78)μm,(6.60±0.25)% vs. (4.27±0.18)%,(4.76±0.33)%;(78.01±18.31)μm vs. (69.98±12.85)μm,(68.45±13.84)μm,(10.62±0.25)% vs. (8.11±1.22)%,(5.89±0.60)%],the difference was statistically significant (P<0.05).Conclusion The glycosyla-tion modification of vimentin could be used for PC12 cell differentiation.
2.Study on gentamicin sulfate regulating gut microbiota for affecting glucose and lipid metabolism and inflammation to improve hypertension in rats

Objective To investigate the relationship between gentamicin sulfate mediated gut microbi-ota in affecting glucolipid metabolism and inflammation with hypertension improvement.Methods Sprague-Dawley rats (8-weeks-old) were divided into the normal diet group (ND,n=11),high fat diet group (HFD,n=13) and HFD combined with gentamicin sulfate gavage group (GS,n=13).Blood pressure was monitored weekly during 5-13 weeks,and the levels of insulin,IL-1β,IL-6,TNF-α,LPS,TC,TG,LDL-C,HDL-C and homeostasis model assessment of insulin resistance index (HOMA-IR) were assessed at 13 weeks.The met-agenomic analyses were performed on DNA samples from rat colonic feces.Results The blood pressure in 13 weeks had no statistical difference among the three groups (P>0.05).The blood pressure during 5-10 weeks in the GS group was higher than that in the ND group with statistical difference (P<0.05).The blood pressure during 5-13 weeks in the HFD group was higher than that in the ND group,and the blood pressure during 10-13 weeks in the GS group was lower than that in the HFD with statistical difference (P<0.05). Compared with the group ND,the levels of TC,TG,LDL-C and HOMA-IR in the HFD group were higher,the HDL-C level was lower,and the differences were statistically significant (P<0.05).Compared with the HFD group,the levels of TG,LDL-C and HOMA-IR in the GS group were lower,the HDL-C level was higher,and the differences were statistically significant (P<0.05).The levels of LPS,IL-1β,IL-6 and TNF-α in the HFD group were higher than those in the ND group,while the GS group was lower than the HFD group (P<0.05).The partial least squares discriminant analysis showed that the intestinal microbiota in the HFD group and GS group was different in structure from that in the ND group.The family level results showed that the structure of the gut microbiota changed in three groups,including 24 microbiotas with significant changes.The hierarchical analysis showed that the proportion of f-desulphovibrio in the HFD group was higher than that in the ND group,while the GS group was lower the HFD group,and the differences were statistically significant (P<0.05).Conclusion Oral antibiotic gentamicin sulfate could improve the glucose and lipid metabolism ab-normalities and inflammation caused by HFD and its mechanism may be to relieve hypertension by regulating intestinal flora structure,abundance and LPS level.
3.Analysis on diagnostic efficiency of interleukin 27,adenosine deaminase and γ-interferon in tuberculous pleurisy

Objective To investigate the diagnostic efficiency of interleukin-27 (IL-27),adenosine de-aminase (ADA) and interferon-γ (IFN-γ) in tuberculous pleurisy (TBP).Methods The pleural effusion sam-ples in 315 cases visiting in this hospital from May 2022 to May 2023 were collected.Among them,165 cases diagnosed as TBP were included in the TBP group,and the other 150 samples were included in the control group (mainly malignant pleural effusion).The levels of IL-27,ADA and IFN-γ in pleural effusion were detec-ted.The receiver operating characteristic (ROC) curve was drawn,and the sensitivity and specificity of each indicator alone and combined diagnosis were evaluated by the area under the ROC curve (AUC).Results The levels of IL-27,ADA and IFN-γ in the TBP group were higher than those in the control group (P<0.05). The AUC value of single indicator in diagnosing TBP all exceeded 0.85,in which AUC of ADA for diagnosing TBP was maximal (0.92),when its optimal cutoff value was 22.48 U/L,the sensitivity and specificity of diag-nosis were 80.61% and 93.33%,respectively,when the combined diagnosis,the diagnostic efficiency of IL-27 and IFN-γ combination was optimal,the AUC value was 0.94,the sensitivity and specificity were 89.70% and 93.33% respectively.Conclusion The single indicator of IL-27,ADA and IFN-γ has the higher diagnostic ef-ficiency for TBP,but the combination diagnosis of IL-27 and IFN-γ has more advantages.
4.Analysis on clinical characteristics and drug resistance of postoperative incision infection caused by Mycoplasma hominis

Objective To analyze the clinical characteristics and drug resistance of postoperative incision infection caused by Mycoplasma hominis (Mh).Methods A total of 8 cases of postoperative MH infection in this hospital from January 2020 to September 2022 were collected,and 8 subjects undergoing the physical ex-amination during the same period were selected as the healthy control group.The data of the disease cases conducted the summarized analysis.Results The Mh caused surgical infection sites were mainly concentrated in the surgical local incision,and the symptoms were manifested as mild redness,swelling,heat,pain and inci-sion poor healing.Moreover,the empirical treatment with β-lactam drugs showed the poor efficacy,and the in-fection cycle lasted for 11-24 d.After obtaining the microbiological evidence,the medication regimen was ad-justed to conduct the target treatment,the incision infection in the patients were effectively improved.Com-pared with the healthy control group,the white blood cell count,neutrophil granulocyte percentage and C reac-tive protein in the infection group were significantly elevated (P<0.01),while the procalcitonin level had no significant change (P>0.05).Mh was cultured in the blood agar medium for 48 h,only small needle-like colo-nies could be seen,moreover the bacteria with Gram stain was not stained.Mh showed good sensitivity to dox-ycycline,minocycline,josamycin,spectinomycin and gatifloxacin,but had high resistance rates to other macrol-ides,aminoglycosides and quinolones.Conclusion Mh caused surgical incision infection is relatively rare.Fully understanding the infection characteristics of this pathogen is beneficial to the diagnosis and treatment of the patients.
5.Construction and application value of a survival prediction model for patients with extramedullary plasmacytoma based on SEER database

Objective To construct a survival prediction model for the patients with extramedullary plasmacytoma (EMP),and to evaluate its application value.Methods The data of the patients diagnosed as EMP from 2000 to 2019 were collected from the National Cancer Institute's Surveillance,Epidemiology,and End Results (SEER) database.Random forest algorithm was used to screen variables,multivariate Cox pro-portional hazards regression model was used to determine the independent risk factors of EMP prognosis,and a nomogram model was constructed based on these factors.The total score of the patients was calculated ac-cording to the nomogram model,and the total samples were divided into the low,medium and high risk groups according to the optimal cutoff value by using X-tile software,and the Kaplan-Meier survival curves of the three groups were analyzed.The predictive performance of the model was evaluated using area under the curve (AUC) of time-dependent receiver operating characteristic (t-ROC) curve,AUC after X years N repeated K-fold cross-validation,calibration curve,and decision curve analysis (DCA).Results A total of 1458 patients with EMP were included,including 796 cases of deletions and 662 cases of death.In the deletion and deaths groups,the majority were aged 60-<75 years old (35.4%,41.2%),male (63.1%,66.3%),white race peo-ple (81.3%,80.4%),and married (68.2%,59.4%).Finally,age (45-<60 years old/60-<75 years old/≥ 75 years old),marital status (single/others),non-primary malignant tumor,and without surgery and radio-therapy were determined to be the independent risk factors for the survival of EPM patients (P<0.05).Based on the above independent risk factors,a survival prediction model was constructed,and a nomogram was drawn.According to the optimal cutoff value,the total sample was divided into the score of 0-96 group (low risk group),score of 98-135 group (medium risk group) and score of 139-191 group (high risk group).The Kaplan-Meier survival analysis revealed that there was statistically significant difference in the survival time a-mong the three groups (P<0.0001).AUC of t-ROC curves for 1,3,5 years and AUC after cross-validation all were>0.7,indicating that the model had a good degree of differentiation.The calibration curves suggested a good consistency between the prediction and practical over survival rate,and DCA indicated that the model could improve the clinical benefits.Conclusion The constructed survival prediction model for EMP patients based on the SEER database could help the clinicians to identify the prognostic risk factors and predict the o-verall survival rate of the patients.
6.Study on relationship between docosahexaenoic acid level and attention deficit hyperactivity disorder based on Mendelian randomization

Objective To investigate the relationship between docosahexaenoic acid (DHA) level and the onset of attention deficit hyperactivity disorder (ADHD) based on Mendelian Randomization (MR).Meth-ods The instrumental variables for DHA and ADHD were extracted from the genome-wide association stud-ies (GWAS) data.To ensure the stablity of the study results,the ADHD datasets were selected from two dif-ferent studies.The The dataset for DHA levels included 115,006 individuals,the ADHD1 dataset consisted of 55374 individuals,including 20183 cases and 35191 controls,the ADHD2 dataset consisted of 225534 indi-viduals,including 38691 cases and 186843 controls,the comprehensive MR and sensitivity analyses were per-formed.The MR analyses were performed by the inverse variance weighted (IVW) method,MR-Egger regression analysis and the weighted median method (WM),utilizing the odds ratio (OR) and effect coefficient (β) as evaluation indictors to explore the bidirectional causal relationship between DHA and ADHD.Results There exists a causal link between DHA levels and the onset of ADHD (ADHD1 IVW:OR=0.833,95%CI:0.719-0.964,P=0.014;ADHD2 IVW:OR=0.895,95%CI:0.808-0.992,P=0.034).Additionally,a reverse causal relation-ship between ADHD and DHA levels (IVW:β=-0.073,95%CI:-0.103 to-0.043,P<0.001) was found.Conclusion The potential bidirectional causal relationship exists between DHA level and ADHD.
7.Expression and significance of BCAS3 and RFX3 protein in macrophages of gout stone tissue

Objective To analyze the expression and significance of BCAS3 and RFX3 proteins in mac-rophages from gout stone tissues.Methods Thirty gout stone excision specimens in this hospital from April 2019 to December 2022 were collected as the experimental group,and during the same period 25 specimens of synovial hyperplasia accompanied by foreign body giant cell reaction after joint replacement and 5 specimens of fat necrosis accompanied by macrophage hyperplasia served as the control group.The blood uric acid level of the control group was normal.Immunohistochemical staining was applied to conduct CD68,CD163,BCAS3 and RFX3 antibodies staining in all specimens.CD68 and CD163 were used to locate macrophages in tissues. The expression levels of BCAS3 and RFX3 in macrophages were detected.The expressions of the two antibod-ies and their correlation with age,sex and blood uric acid level were analyzed.The receiver operating character-istic (ROC) curve was used to evaluate the diagnostic value of the both on gout.Results The expression lev-els of BCAS3 and RFX3 in the experimental group were significantly higher than those in the control group (P<0.05).The BCAS3 and RFX3 protein expression levels in the control group had no statistical difference between the different sexes (P>0.05).The expression level of BCAS3 protein in the experimental group was positively correlated with the RFX3 protein expression (P<0.05);the uric acid level in the experimental group was positively correlated with the expression level of BCAS3 and RFX3 protein (P<0.05).The blood uric acid level in the control group had no correlation with BCAS3 and RFX3 protein (P>0.05).The age in the two groups had no correlation with the BCAS3 and RFX3 protein (P>0.05).The area under the curve (AUC),sensitivity and specificity of RFX3 were 0.935,96.55% and 86.67% respectively,which were higher than 0.742,70.00% and 80.00% of BCAS3.RFX3 had the higher diagnostic value for gout.Conclusion High expression of BCAS3 and RFX3 proteins in macrophages may participate in the pathogenesis of gout.There may be some synergistic promoting effect between the two.The diagnostic value of RFX3 for gout is higher than that of BCAS3.
8.Effects of mindfulness-based parenting course on parenting burnout and parent-child relationship of adolescents' mothers

Objective To investigate the effects of mindfulness-based parenting course on parenting burnout and parent-child relationship of adolescents' mothers.Methods A total of 203 mothers who volun-teered to participate in the mindfulness-based parenting course in a middle school in Chongqing City in May 2023 were recruited as the research subjects and divided into the intervention group (n=99) and control group (n=104).The intervention group attended the eight mindfulness parenting sessions twice a week,while the control group did not attended the course.The Parenting Burnout Assessment Scale,Parent-Child Relationship Self Evaluation Scale,Five Factors of Mindfulness scale and Satisfaction with Life Scale were used to evaluate the two groups before and after intervention.Results The scores of parent-child relationship and satisfaction with life had statistical difference between the two groups (P<0.05).Compared with before intervention,the parental burnout score of the intervention group was decreased,the scores of parent-child relationship,five fac-tors of mindfulness,scores of awareness,no judgment,no reaction dimensions and life satisfaction were in-creased,the differences were statistically significant (P<0.05).Compared with the control group,the scores of parent-child relationship,five factors of mindfulness,awareness dimension and life satisfaction after inter-vention in the intervention group were higher,while the score of conscious action dimension was lower,the differences were statistically significant (P<0.05).Conclusion The 4-week mindfulness-based parenting course could could decrease the parenting burnout,and improve the parent-child relationship and satisfaction with life.
9.Clinical features of invasive Streptococcus pneumoniae disease in children and early warning indicators of severe case

Objective To investigate the clinical features in children with invasive pneumococcal disease (IPD) and early warning indicators of severe case.Methods The case data of 101 children inpatients with IPD isolated Streptococcus pneumoniae (Sp) at least once blood culture in this hospital from January 2007 to De-cember 2021 were collected retrospectively.The children patients were divided into the pediatric intensive care unit (PICU) group (35 cases) and general ward group (66 cases) according to whether or not entering PICU during hospitalization.The general clinical data,infection types and results of drug sensitivity test and bacteri-al strain serotype identification were compared between the two groups and analyzed.Results Compared with the general ward group,the male proportion,proportions of complicating nervous system disease and iron-defi-ciency anemia,winter incidence rate,disturbance of consciousness,convulsion,dyspnea,heart rate increase,oc-currence rate of vomiting and diarrhea,proportion of peripheral blood neutrophilic granulocytes,C reactive protein (CRP),lactic dehydrogenase (LDH) level,Hb<90 g/L,LDH>700 U/L,creatine kinase isoenzyme-MB (CK-MB) abnomality,proportions of bacterial meningitis,sepsis and severe pneumonia in the PICU group were higher,the proportion of 3-97 percentile of body weight,bronchopneumonia and no-focus blood stream infection were lower,WBC,Hb and albumin level were lower,and the differences were statistically significant (P<0.05).Eleven kinds of serotypes were identified in this study,ranking the top five in order were 6B,14,19F,23F and 19A.Pneumococcal 13-valent conjugate vaccine (PCV13) could cover 96.9% of the IPD sero-types.The multivariate logistic regression analysis showed that consciousness disturbance,convulsion,per-centage of neutrophils increase,Hb<90 g/L,LDH>700 U/L and CK-MB abnormality indicated the condition of IPD children patients was critical,which was the early-warning indicator of entering PICU (P<0.05).Con-clusion Severe IPD lacks the early specific early-warning indicator,and the cover rate of PCV13 is high.
10.Analysis on risk factors of severe acute pancreatitis complicating splanchnic venous thrombosis

Objective To investigate the risk factors of complicating splanchnic venous thrombosis (SVT) in severe acute pancreatitis (SAP) in order to guide clinic for conducting early prevention.Methods A total of 173 patients with SAP admitted and treated in this hospital from January 2018 to December 2022 were selected and divided into the non-complicating SVT group (n=143) and complicating SVT group (n=30) ac-cording to whether having secondary SVT or not.The general data and various clinical treatment and examina-tion-related data were collected.The multi-factor logistic regression was used to analyze the risk factors of complicating SVT in SAP.Results Shock (P<0.001),pancreatic cyst (P=0.002),tracheal intubation (P<0.001),proportion of abnormal renal function (P=0.014),total bilirubin (P=0.049),direct bilirubin (P=0.018),AST (P=0.011),procalcitonin (P=0.021),thrombin time (P=0.024),D-dimer (P=0.003),MCTSI score (P<0.001) and APACHE Ⅱ score (P<0.001) had statistical difference between the two groups.The multivariate logistic regression analysis showed that pancreatic cyst (OR=2.700,95%CI:1.022-7.134,P=0.045) and APACHE Ⅱ score increase (OR=1.137,95%CI:1.002-1.290,P=0.047) were the independent risk factors for complicating SVT in SAP.Conclusion Pancreatic cysts and APACHE Ⅱ score increase are the independent risk factors for complicating SVT in SAP.These patients require close fol-low-up and timely intervention and treatment.
